چکیده

Background on the Chemistry of Rotenone. Rotenone is a natural product isolated from certain subtropical and tropical members of the pea family found in South America and Southeast Asia. The use of rotenone by man may be ancient, as early explorers noted Peruvian natives using crude extracts of the Cubé plant to stun fish for eating. In the early 1900’s, botanists exploring Peruvian jungles searching for new and useful plants of potential commercial value exported large quantities of plant roots and extracts to the US for use as insecticides on food crops, with the major active ingredient of the extracts being rotenone. Rotenone has been used commercially as a garden insecticide since the middle 1800’s, primarily in the UK. Rotenone is still currently widely used as a garden insecticide, and in organic farming as a natural, botanical insecticide. In California, about 200 kg of rotenone is used per year as an insecticide for organically grown lettuce and tomatoes. Rotenone has also found significant use as a highly selective piscicide for the control or eradication of non-native fish in streams and lakes across the United States. Rotenone has been used successfully for native trout reintroduction in the United States since the 1960’s.

The effect of low dose amphetamine in rotenone-induced toxicity in a mice model of Parkinson’s disease

Objective(s): The effects of low dose amphetamine on oxidative stress and rotenone-induced neurotoxicity and liver injury were examined in vivo in a mice model of Parkinson’s disease. Materials and Methods: Male mice were treated with rotenone (1.5 mg/kg, every other day for two weeks, subcutaneously). Mice received either the vehicle or...

Fate and behavior of rotenone in Diamond Lake, Oregon, USA following invasive tui chub eradication.

In September 2006, Diamond Lake (OR, USA) was treated by the Oregon Department of Fish and Wildlife with a mixture of powdered and liquid rotenone in the successful eradication of invasive tui chub Gila bicolor. During treatment, the lake was in the middle of a phytoplankton (including cyanobacteria Anabaena sp.) bloom, resulting in an elevated pH of 9.7. Dissipation of rotenone and its major m...

Selective vulnerability of dopaminergic neurons to microtubule depolymerization.

Parkinson disease (PD) is characterized by the specific degeneration of dopaminergic (DA) neurons in substantia nigra and has been linked to a variety of environmental and genetic factors. Rotenone, an environmental PD toxin, exhibited much greater toxicity to DA neurons in midbrain neuronal cultures than to non-DA neurons. The effect was significantly decreased by the microtubule-stabilizing d...

Neurotrophic factors stabilize microtubules and protect against rotenone toxicity on dopaminergic neurons.

Parkinson disease is characterized by the selective degeneration of dopaminergic (DA) neurons in substantia nigra. Long term epidemiological studies have implicated exposure to agricultural pesticides as a significant risk factor. Systemic administration of rotenone, a widely used pesticide, causes selective degeneration of nigral DA neurons and Parkinson disease-like symptoms in rats. Our prev...

Rotenone selectively kills serotonergic neurons through a microtubule-dependent mechanism.

As a major co-morbidity of Parkinson's disease (PD), depression is associated with the loss of serotonergic neurons. Our recent study has shown that midbrain dopaminergic neurons are particularly vulnerable to microtubule-depolymerizing agents including rotenone, an environmental toxin linked to PD. Here we show that rotenone also selectively killed serotonergic neurons in midbrain neuronal cul...
